Briar gains the charge counter even if the card played is negated or destroyed in some way.
Causing Briar to lose her abilities will not make her lose any charge counters she has accrued.
If the ability is negated by having Briar lose her abilities through an effect like Silence, then you will still have to pay the mana (M) cost AND remove the charge counter.
